Показано участие ФНО в меха­низме таких заболеваниях, как болезнь Альцгеймера, Паркинсона и рассеянный склероз. Тем не менее роль ФНО в ЦНС в нормальных физиологических условиях изучена слабо. В Институте молекуляр­ной биологии им. В.А. Энгельгардта РАН группой С.А. Недоспа­сова была создана линия мышей с нокаутом по ФНО (TNFKO) на основе генома линии C57Bl/6 (WT). В нашей работе мы сравнивали поведение половозрелых самцов TNFKO и WT в стандартной батарее тестов, включающей «открытое поле», «приподнятый крестообразный лабиринт» и «принудительное плавание». Мы показали, что дефицит ФНО не влияет на двигательную активность и исследовательское поведение мышей в тесте «открытое поле». Тем не менее в этом тесте мыши TNFKO больше времени проводили в центре арены по сравнению с мышами WT, но при этом имели выше уровень дефекаций и ниже длительность стоек. Этот результат свидетельствует о том, что легкий стресс в тесте «открытое поле» скорее дезориентирует мышей TNFKO, чем вызывает у них тревогу. Не было обнаружено различий по выраженности тревожности в тесте «приподнятый крестообразный лабиринт» и депрессивно-подобного поведе- ния в тесте «принудительное плавание» между мышами этих линий. Полученные данные позволяют предположить, что отсут­ствие данного цитокина влечет за собой изменения в нейрофункциональных взаимодействиях, что приводит к изменению у мышей ответа на легкий стресс в тесте «открытое поле».</p></abstract><trans-abstract xml:lang="en"><p>The tumor necrosis factor (TNF) is a cytokine exerting both homeostatic and pathophysiological roles in the central nervous system (CNS). It has been demonstrated that TNF plays roles in such diseases as Alzheimer’s disease, Parkinson’s disease and multiple sclerosis. Nevertheless, the role of TNF in the CNS under normal physiological conditions is poorly studied. A novel mouse strain with TNF deficiency (TNFKO) was developed in S.A. Nedospa­sov’s labora­tory of Engelhardt Institute of Molecular Bio­logy of the Russian Academy of Sciences on the C57Bl/6 (WT) background. In our study, we compared the behavior of TNFKO and WT adult mice in a battery of tests: open-field, elevated plus-maze and the forced-swim test. We showed that TNF deficiency had no effect on locomotor activity or exploration in the openfield test. At the same time, in this test, TNFKO mice spent more time in the center of the arena, but had a higher level of defecation and lower rearing duration. This result indicates that, in the openfield conditions, TNFKO mice show disorientation rather than anxiety-like behavior. There were no differences between TNFKO and WT in anxiety level in the elevated plus-maze test or in depressive-like behavior in the forcedswim test.
