Эта болезнь приводит к таким серьезным нарушениям, как инфаркт миокарда, инсульт. Благодаря развитию спектроскопии ядерно-магнитного резонанса показано, что у людей с артериальной гипертензией наблюдается снижение жизнеспособности нейронов различных отделов головного мозга. Трансляция в клиническую практику методов спектроскопии ядерно-магнитного резонанса требует накопления эмпирических данных об изменениях нейрометаболизма в условиях строго контролируемого эксперимента. В частности, представляет интерес сравнение метаболомных параметров у лабораторных животных с нормальным и повышенным артериальным давлением, содержащихся в стандартных условиях, на одинаковой диете. В представленной работе исследованы метаболиты коры головного мозга и гипоталамуса самцов крыс гипертензивной линии НИСАГ (ISIAH) и нормотензивной линии Wistar в возрасте 8–9 нед. У животных, находящихся под газовым наркозом (изофлюран), проводили методом протонной спектроскопии ядерно-магнитного резонанса (1Н МРС) измерение метаболитов в коре головного мозга и гипоталамусе. Обработка первичных данных с помощью анализа Partial Least Squares Discriminant Analysis (PLS-DA) позволила выделить основную дискриминирующую ось (Y1), вариации которой отражали преобладание возбуждающих нейрометаболитов (глутамина и глутамата) над тормозными (ГАМК и глицином). Значения оси Y1 в коре головного мозга были выше у крыс линии Wistar по сравнению с линией НИСАГ, что указывает на снижение возбудимости коры головного мозга у гипертензивных животных. В гипоталамусе, наоборот, величина оси Y1 была выше у крыс линии НИСАГ, чем у крыс линии Wistar. При этом преобладание возбуждающих нейрометаболитов положительно коррелировало с уровнем среднего артериального давления, что хорошо согласуется с представлением об активации каудальных отделов гипоталамуса у гипертензивных животных.</p></abstract><trans-abstract xml:lang="en"><p>Hypertension is one of the most common human diseases. This disease leads to serious disturbances such as myocardial infarction and stroke. Due to the development of nuclear magnetic resonance spectroscopy (NMRS), a decrease in neuron viability in different parts of the brain in humans with hypertension has been shown. Translation of NMRS tools to the clinic requires the accumulation of empirical data about neurometabolic changes in a strictly controlled experiment. It is particularly interesting to compare the metabolic parameters of laboratory animals with normal and high blood pressure kept in standard conditions on exactly the same diet. In this study, cortex and hypothalamus metabolites of ISIAH and Wistar male rats at the age of 8–9 weeks were examined. Cortex and hypothalamus metabolites were measured in animals under isoflurane anesthesia using proton magnetic resonance spectroscopy (1Н MRS). Processing of primary data using Partial least squares Discriminant Analysis (PLS-DA) allowed us to identify the main discriminating axis (Y1), its variations reflecting the predominance of excitatory neurometabolites (glutamine and glutamate) over inhibitory ones (GABA and glycine). In the cortex, the values of the Y1-axis were lower in ISIAH than in Wistar rats. This fact indicates a decrease in cortical excitability in hypertensive animals.
