Известно, что некоторые однонуклеотидные замены (SNP – single nucleotide polymorphism) в ТАТА-боксе и его фланках ассоциируются с повышенным риском возникновения различных патологий человека (Савинкова и др., 2009). Данные об изменении взаимодействия ТВР/ТАТА, приводящем к повышенному риску возникновения какой-либо патологии, в литературе практически отсутствуют. В этой работе изучали влияние SNP (–21t &gt; c), прилегающего к ТАТА-боксу промотора гена NOS2A, на взаимодействие с ТВР человека с использованием метода поверхностного плазмонного резонанса (SPR) на биосенсоре ProteOn XPR36. Из полученных сенсограмм видно, что замена 21t&gt;c в последовательности, прилегающей к ТАТА-боксу, практически не влияла на константу скорости ассоциации ТВР с ТАТА-содержащим олигонуклеотидом, а константы реакции диссоциации отличались примерно в два раза. Таким образом, эксперименты, проведенные с использованием метода SPR и классического метода EMSA, показали, что при иммобилизации ТВР на декстрановой поверхности микрочипа не происходит изменения аффинности его взаимодействия с ТАТА-содержащими олигонуклеотидами.</p></abstract><trans-abstract xml:lang="en"><p>Interaction ofthe TATA-binding protein (ТВР) with the TATA box and formation ofthe TBP/TATA complex is the first step in the transcription complex assembly and the central event in transcription regulation. It is known that some single nucleotide polymorphisms (SNPs) in the human TATA box and its flanks are associated with elevated risk ofvarious disorders (Savinkova etal., 2009). Little is known about changes in the TBP/TATA interaction leading to the elevated risk ofany disease. Thus, we have studied the influence ofSNP (–21 t &gt; c), adjoining to the TATA box ofthe NOS2A promoter, on the interaction by surface plasmon resonance (SPR) on biosensor ProteOn XPR36 (BioRad). The sensograms obtained in the experiment indicate that the constant for the interaction with TATA bearing SNP –21 t &gt; c differ little from the constant with the wild-type box and the dissociation constants differ approximately twofold.
